Herbicide and Plastoquinone Binding to Photosystem II

چکیده
Photoaffinity Label, Plastoquinone Depletion, Photosystem II Particles, Chlamydomonas In plastoquinone-depleted thylakoids “D C M U -type” herbicides generally have a lower binding affinity as compared to control thylakoids, with the exception o f DC M U itself. Contrary, binding properties of phenolic herbicides are not affected by plastoquinone depletion. DCM U in plastoquinone-depleted thylakoids can be displaced from the membrane by various substituted 1,4-benzoquinones, including plastoquinone-1. Labeling patterns by azido-atrazine are compared for thylakoids, plastoquinone-depleted thylakoids, and different photosystem II preparations from spinach and Chlamydomonas. Azido-atrazine tags a 3 2 3 4 kDa protein, which seems to be different from another 3 2 -3 4 kDa protein which is preferentially labeled by a plastoquinoneazide.

The Topology of the Plastoquinone and Herbicide Binding Peptides of Photosystem II in the Thylakoid Membrane
The 32 kDa herbicide and O b binding peptide (D-l protein) and its homologous 34 kDa peptide (D-2 protein) are integral membrane subunits of photosystem II. A model for their folding through the thylakoid membrane in five transmembrane a-helices is proposed from the compari son of amino acid sequence and hydropathy index plot homologies with subunits of the bacterial system. Following recent d...
متن کاملMutations in the D1 Subunit of Photosystem II between Quinone and Herbicide Binding Sites Distinguish
The structure-activity relationships of the plastoquinone Q B binding domain in the D1 subunit of photosystem II (PSII) were investigated by characterization of mutations introduced in the D1 protein. Eight nove1 point mutations in the gene psbA, which encodes D1, were generated in the cyanobacterium Synechocyslis PCC6803 by site-specific mutagenesis in vitro. The effects of the resulting modif...
متن کاملStructural basis of cyanobacterial photosystem II Inhibition by the herbicide terbutryn.
Herbicides that target photosystem II (PSII) compete with the native electron acceptor plastoquinone for binding at the Q(B) site in the D1 subunit and thus block the electron transfer from Q(A) to Q(B). Here, we present the first crystal structure of PSII with a bound herbicide at a resolution of 3.2 Å. The crystallized PSII core complexes were isolated from the thermophilic cyanobacterium The...
متن کاملCorrection: Evidence for the Involvement of Loosely Bound Plastosemiquinones in Superoxide Anion Radical Production in Photosystem II
Recent evidence has indicated the presence of novel plastoquinone-binding sites, QC and QD, in photosystem II (PSII). Here, we investigated the potential involvement of loosely bound plastosemiquinones in superoxide anion radical (O2-) formation in spinach PSII membranes using electron paramagnetic resonance (EPR) spin-trapping spectroscopy. Illumination of PSII membranes in the presence of the...
متن کاملInteractions of Herbicides with Photosynthetic Electron Transport
The two primary sites of herbicide action in photosynthetic electron transport are the inhibition of photosystem II (PS II) electron transport and diversion of electron flow through photosystem I (PS I). PS II electron transport inhibitors bind to the DI protein of the PS II reaction center, thus blocking electron transfer to plastoquinone. Inhibition of PS II electron transport prevents the co...
